I started many years ago with Something Positive. Quickly through its series of links I discovered many others and now i read probably close to fifty a day. Here are just a few of my favorites. Anyone have any new suggestions?

Buttersafe - Punch monster and Skeleton Harvester are awesome
WTF Comics - Everquest-based and not updated very often lately

D&D based: (I've never actually played but have a good enough grasp on the rules that these are still funny)
Order of the Stick (Server is slow sometimes)
Darths and Droids - What if Star Wars were a D&D campaign?

A few more of my favorites that have yet to be mentioned.

Lovecraft is Missing: I love HP Lovecraft and story-driven comics, this is combining both so far.
Looking for Group: From the guys who do Least I could Do, a comic that started out as a WOW parody and has become a pretty fun fantasy comic.
A Softer World: A weird, photo based comic. Pretty rad and alternately hilarious or a little depressing.
Diesel Sweeties: R. Stevens is rad and this is brilliant. A pixel based comic that actually works pretty well.
Overcompensating: One of the reigning kings of webcomics. He also created Wigu which is equally awesome, truly bizarre and really really long, but worth reading through the entire archives.

I do love Dresden Codak too though I don't think I'm supposed to. I'm pretty sure its the most popular webcomic to hate on, right behind Ctrl+Alt+Del.

I like Perfect Stars too. The art is amazing.

I can't believe Schlock Mercenary hasn't been mentioned here. Take equal parts hard sci-fi, epic adventure, weapons grade puns, and a title character who resembles a pile of crap, and you get 100% pure awesome. You should really start from the beginning, though.
